The agreement covers around 100 workers at the museum in the Clerkenwell area of central London.
The union said workplace problems, including the lack of a proper cooling system last year, had highlighted the benefits of a collective voice and helped drive the campaign.
Prospect deputy general secretary Noel McClean said: “As a former postie, this is an area that is close to my heart, so I am extremely pleased to have signed this agreement.
“It means workers at the Postal Museum now have access to all the benefits a collective voice brings.”
Postal Museum chief executive Laura Wright said: “We’re pleased to formally recognise Prospect.
“The Postal Museum is committed to fostering an inclusive and welcoming culture where colleagues feel respected, listened to and able to participate in shaping their working lives.
“Open dialogue and a recognised collective voice are important to that ambition.”
